An independent laboratory's method being different from the supplier's is not a discrepancy. It becomes one only when the results differ by more than both methods' demonstrated precision.

Worth one more sentence than it usually gets.

Building on post #30 rather than restating it.

Adding a reference point for Forced degradation studies. Mine is a single case, collected without controls, and I am posting the method alongside it so it can be discounted appropriately.

t.wojcik, post #12: Worth separating two things that post #10 runs together. Specificity: the method can distinguish the intended compound from related impurities and degradation products. Tested by comparing results on pure compounds, mixtures of compounds, and degraded samples. Go to post

Transfer between laboratories: a method can be transferred from one lab to another, but the receiving lab needs to demonstrate that they can achieve the same performance. This requires comparative testing and sometimes small method refinements.

Why two laboratories may disagree: after validating the same method, they may still report different purity on the same sample due to integration differences, column age differences, subtle differences in mobile phase pH or temperature. This is normal and not a sign that one is wrong.

That is a description of practice, not a recommendation of it.

The version of Forced degradation studies that circulates here is a simplification of a simplification. It is not wrong, but it has lost the conditions under which it holds, and those conditions are where the interesting cases live.

Where I part company with post #36, and it is a narrow parting.

Something worth flagging about Forced degradation studies: the strongest-sounding claims in this thread are the ones with no source attached, which is the usual pattern and not a coincidence.

a.kowalski, post #17: Post #15 is right about the mechanism and I think understates the practical bit. Adding a small correction to the Forced degradation studies summary above rather than a disagreement with it. The substance holds; one of the figures is out by a factor that matters. Go to post

I have three months of notes on Forced degradation studies and the honest summary is that the trend is real and the week-to-week numbers are noise. I nearly drew the opposite conclusion from the first fortnight.
