A request rather than an answer: could whoever has the primary source for Mean kinetic temperature post it? I have seen the claim three times this month and each version had lost a qualifier.

Revisiting: Mean kinetic temperature explained without the mathematics Writing it up because I had to work it out twice and would rather nobody else did.

A methods question rather than a substantive one, about Mean kinetic temperature.

Everyone quotes the same figure and I cannot find anyone who says how it was arrived at. That is not an accusation; it usually means the derivation is somewhere obvious and I have missed it.

The useful distinction on Mean kinetic temperature is between what was measured and what was inferred from it. Both end up in the same sentence and only one of them has error bars.

I read the opening post twice before replying, because I had assumed the opposite.

Clarity is a weak test. A solution can lose a significant fraction of its active content to degradation or adsorption while remaining perfectly clear.

If anyone has run this properly I would rather read that than my own guess.

Two claims get bundled together under Mean kinetic temperature and they need separating. The descriptive one — this is what was observed — is usually well supported. The causal one — this is why — usually is not.

Almost every disagreement in threads like this one dissolves once you say which of the two you are making.

Visible particulate is a strong signal in the other direction: it is a reason to stop and ask rather than to filter and proceed.

Not a strong opinion, just a consistent one.

What would change my mind on Mean kinetic temperature is a second dataset collected by someone with no stake in the first. Until then I hold it loosely and I would rather say so than pretend to more.

Temperature excursions in transit are common and are not all equal. A few hours warm is a different event from three days warm, and neither is visible in the vial on arrival.

Temperature excursions: if a vial sat at room temperature for a time, the question is whether the excursion was long enough to cause damage. Hours at room temperature is usually low risk. Days to weeks raises the risk. If you are unsure, contacting the supplier is more informative than guessing.

If anyone can point at the primary source I would be grateful.

Light protection is easy and cheap. The original carton exists for a reason and a vial stored in it is protected without anybody having to think about it again.

Reconstituted material carried in a bag for a day is a temperature excursion even in a cool climate. Where that matters is cumulative exposure over weeks rather than a single afternoon.

That is the practical version. The rigorous version is longer and says the same thing.

Post #16 and I disagree about the size of the effect, not about the direction.

Mean kinetic temperature: I would want to see the raw numbers rather than the summary before agreeing. Summaries lose exactly the information that would settle this.

Adsorption to container surfaces removes peptide from solution without changing how the solution looks. In dilute preparations that loss can be a meaningful proportion, and it is invisible.

Two sources, same conclusion, and I could not rule out that one copied the other.

Two questions I would want answered before drawing anything from the Mean kinetic temperature data above: how were the cases selected, and what happened to the ones that dropped out.

Coming back to post #21, because the follow-up matters more than the original answer.

I would keep Mean kinetic temperature and the decision it usually gets used for separate in this thread. They are related and they are not the same question, and merging them is why the last one went badly.

Taking post #24 at face value and following it one step further.

The honest summary: keep it dry until you need it, keep it cold once it is not, keep it dark, do not cycle the temperature, and treat any period at room temperature as spent whether or not anything visible happened.
