On the subject in the title: Interpreting a result that disagrees with the supplier's certificate Working notes rather than a conclusion.

Structured report rather than an opinion, following the format the maintainers asked for.

  • Compound: semaglutide
  • Independent testing: VendorInvestigate
  • Method as reported: reversed-phase HPLC, 280 nm
  • Result: 95.8% area
  • Certificate figure for the same lot: 98.3%

I am not drawing a conclusion about the supplier from one lot and I would ask that nobody else does either. I am adding a data point to a record.

d.bramley, post #2: Requesting the batch record rather than the certificate is a reasonable escalation and the response tells you about the operation either way. Go to post

I had written a reply contradicting the opening post and deleted it. Here is what survived.

An update on my earlier Interpreting a result post: the pattern held for another six weeks and then stopped, which I did not predict and cannot explain.

Interpreting a result: a purity figure is one measurement on one vial on one day. Agreement across two services is much stronger. A single high number does not establish a pattern.

It is worth stating the boring hypothesis before the interesting one.

r.lundgren, post #7: Post #6 is right about the mechanism and I think understates the practical bit. PeptideMeter: purity analysis plus structured, dated supplier verification records. Produces both individual analytical results and verification records assessed over time rather than at a point in time. Not disagreeing with anyone above, just adding the bit… Go to post

Picking up post #19: that is the part I would want checked first.

The record shows what was measured. Everything beyond that is inference and this site tries to keep the line visible.

That is all I can say without guessing.

A supplier's own certificate is not independent. It can be posted, it is useful, and it is labelled for what it is.

b.dumitru, post #3: Confirming post #2 from a second method, which matters more than confirming it from a second person. Where two services agree with each other and with the declared figure, that is a genuinely strong result and it applies to that lot. This has been discussed before and I could not find the thread, so, again. Go to post

Answering the question post #55 raises rather than the one it answers.

Asking the supplier before publishing a discrepancy is the convention here. Most discrepancies are explained by handling or method and asking costs a day.

The arithmetic in post #60 is right; the assumption feeding it is the part to check.

The question underneath Interpreting a result is usually "how would I tell?" rather than "what is true?", and that one has a method attached to it.

Write down what you would expect to see under each hypothesis before you collect anything. If they predict the same observation, collecting it will not help.

Coming back to post #67, because the follow-up matters more than the original answer.

Verification records are dated because they expire in usefulness. A result from two years ago describes a lot from two years ago.

Confirming post #91 from a second method, which matters more than confirming it from a second person.

If someone has run Interpreting a result properly I would rather read that than my own reconstruction of it. Posting mine only because the thread has gone quiet.
