Thibodeau, post #2: Beyond-use dating: the date after which a prepared solution should not be used. It is an estimate based on how fast the solution degrades under stated storage, not on whether the solution suddenly becomes unusable on a calendar date. The date is a boundary; crossing it is a risk factor not a certainty of damage. Go to post

Clarity is a weak test. A solution can lose a significant fraction of its active content to degradation or adsorption while remaining perfectly clear.

Where I part company with post #60, and it is a narrow parting.

Vial integrity: once a vial is entered with a needle repeatedly, the rubber closure degrades and the integrity fails. Using the same vial repeatedly over weeks or months carries increasing risk of contamination. Single-use vials entered once are lowest risk.

What I would check first on reconstituted is whether the thing being measured moved or whether the way of measuring it moved. Those look identical in a graph.

Filing a mild objection to the consensus on reconstituted. Mild because I might be wrong; an objection because nobody has addressed the case that does not fit.

m.yilmaz, post #59: Container choice matters: the material of the vial, the rubber closure, the fill volume all affect how quickly the contents degrade. Smaller fill volumes lose less to adsorption. Glass vials lose less to leaching than plastics. Go to post

Light protection is easy and cheap. The original carton exists for a reason and a vial stored in it is protected without anybody having to think about it again.

I have left out the parts I could not verify.

I had written a reply contradicting post #64 and deleted it. Here is what survived.

Reconstituted material carried in a bag for a day is a temperature excursion even in a cool climate. Where that matters is cumulative exposure over weeks rather than a single afternoon.

Check the ordinary explanations, in order, and stop when one of them accounts for what you are seeing. Most of the time the second one does. Go to post

The claim about reconstituted upthread is stronger than its source supports. I have read the source. The source says "associated with" and the post says "causes".

Temperature excursions: if a vial sat at room temperature for a time, the question is whether the excursion was long enough to cause damage. Hours at room temperature is usually low risk. Days to weeks raises the risk. If you are unsure, contacting the supplier is more informative than guessing.

Published stability data applies specifically to the formulation studied, under the specific conditions stated. It does not automatically transfer to something reconstituted at home in a different concentration with a different diluent. That caveat is not small.

The answer changed when I changed how I was measuring, which was informative.

Freeze-thaw cycling is one of the harsher things you can do to a protein in solution, and a single freeze is different from repeated ones. I am not the right person to answer the follow-up to this. The failure mode on reconstituted is boring rather than dramatic. It is almost always the step everyone assumes was done correctly because it is too simple to get wrong.

Adding a reference point for reconstituted. Mine is a single case, collected without controls, and I am posting the method alongside it so it can be discounted appropriately.

Dating prepared solutions: write the date reconstitution on the vial in permanent marker. If you later need to know whether it has been sitting for weeks or days, the date tells you. Guessing on this is where errors accumulate.

A single observation, in a thread that deserves better than single observations.

A few hours warm is a different event from three days warm, and neither is visible in the vial on arrival. That is the practical version. The rigorous version is longer and says the same thing. Building on post #79 rather than restating it.

A methods point on reconstituted rather than a substantive one: if the comparison is not like for like, the difference you are measuring is the difference in method.

Confirming post #80 from a second method, which matters more than confirming it from a second person.

A beyond-use date on a home preparation is an inference rather than a measurement. It can be a reasonable inference and it should be described as one.

Somebody will have a better source than mine, and I hope they post it.

On reconstituted, I would rather understate and be corrected upward than overstate and be quoted. That is a house style here and it is a good one.

One caution on reconstituted: everything above assumes the underlying documentation is what it claims to be. That assumption is doing real work and is rarely stated.
