Reconstitution volumes in this family are often small enough that dead volume in the syringe is a meaningful fraction of the dose. A fixed-needle insulin syringe is worth the trouble here specifically.

Confirming post #60 from a second method, which matters more than confirming it from a second person.

CJC-1295 is well covered in the tag pages, and the older discussions are better than the recent ones because they were argued out properly. Worth twenty minutes before adding to this one.

I had written a reply contradicting post #62 and deleted it. Here is what survived.

Effects reported at the level of appetite are the most consistently described and the most mechanistically direct for the ghrelin-receptor compounds, which is not what most people are taking them for.

I am reporting what happened, not recommending it.

Reading back through the CJC-1295 threads from last year, the same three questions come up every time and only one of them has ever been answered properly. That seems like a documentation gap rather than a knowledge gap.

Nobody has said the unglamorous part of CJC-1295 yet, so: most of the variation is explained by things that are boring to write about and easy to check.

Answering the question post #66 raises rather than the one it answers.

GHRH analogues versus ghrelin mimetics are often conflated but they are different mechanisms. GHRH analogues (like CJC-1295) cause the pituitary to release growth hormone. Ghrelin mimetics (like GHRPs) act on the ghrelin receptor directly. The effect on growth hormone secretion is similar but the mechanisms are distinct.

On analysis: these are short peptides and generally straightforward chromatographically, which means a poor purity result is more likely to reflect the synthesis than the method.

That is one dataset and I would not build a rule on it.

The number people quote for CJC-1295 is a central estimate presented without its interval, and the interval is wide enough that the estimate is nearly uninformative on its own.

Knowlton, post #17: Injection-site reactions are reported more often in this family than for the incretins. Whether that is the compounds or the preparations they are typically supplied in is not established. I am not the right person to answer the follow-up to this. Go to post

Reading a rodent study on a secretagogue without over-extrapolating: rodent studies can show a mechanism is plausible. They cannot show magnitude of effect in humans or safety profile in humans. That gap is larger for secretagogues than for incretin agonists because the human evidence is thinner.

Picking up post #73: that is the part I would want checked first.

The version of CJC-1295 that I was taught turned out to be a teaching simplification. Useful, and not true in the way I had assumed it was.

One caution on CJC-1295: everything above assumes the underlying documentation is what it claims to be. That assumption is doing real work and is rarely stated.

CJC-1295 with and without DAC: the version with a drug affinity complex binds albumin covalently and persists for days. The version without it does not persist. Supplier labelling frequently does not distinguish them clearly, so confirming which you have is important.

Worth separating two things that post #73 runs together.

CJC-1295 is a good example of a question where the honest answer is boring and the interesting answers are unsupported. I would go with boring.

Post #80 put the caveat in the right place and I want to underline it.

Two sentences on CJC-1295 and then I will stop, because the rest is speculation and the thread is better without mine.

What is documented is narrow. What is inferred from it is broad. The gap between them is where every argument here lives.

A definition problem is doing most of the work in this CJC-1295 discussion. Once the term is pinned down I suspect the disagreement mostly goes away and what is left is small.

The claim about CJC-1295 upthread is stronger than its source supports. I have read the source. The source says "associated with" and the post says "causes".

Post #84 and I disagree about the size of the effect, not about the direction.

A mass result for a short peptide is more discriminating than for a long one, because a single residue difference is a larger proportion of the total. That makes identity confirmation genuinely useful here.

Noting that I have skin in this question and have tried to discount for it.
