On the subject in the title: GHRH analogues versus ghrelin mimetics: different mechanisms, conflated discussion Working notes rather than a conclusion.

Asking about GHRH analogues versus ghrelin mimetics directly, because I have read four threads on it and each answered a slightly different question.

The version I want answered is the narrow one: given the method stated below, is the result within what anyone else has seen? I am not asking what it means yet.

Method, numbers and the two assumptions I am aware of making are below. If the assumptions are wrong that is more useful to me than agreement.

CJC-1295 exists in two forms in circulation — with and without the drug affinity complex — and they have very different half-lives. Product descriptions frequently do not say which, and the difference is not cosmetic.

Trying to state the GHRH analogues versus ghrelin mimetics position in a way that someone who disagrees would recognise as fair, because I do not think the version in this thread passes that test.

A mass result for a short peptide is more discriminating than for a long one, because a single residue difference is a larger proportion of the total. That makes identity confirmation genuinely useful here.

Ipamorelin selectivity claims originate in preclinical characterisation and are reasonably well supported for what they claim: less effect on cortisol and prolactin than earlier GHRPs. Whether that translates to something clinically meaningful is a different question from whether the selectivity is real.

That holds under the stated conditions and I have stated them.

Injection-site reactions are reported more often in this family than for the incretins. Whether that is the compounds or the preparations they are typically supplied in is not established.

The honest answer is that it depends, and here is what it depends on.

Adding a null result on GHRH analogues versus ghrelin mimetics. I looked, carefully, and found nothing, and null results deserve posting precisely because they never are.

Agreed on GHRH analogues versus ghrelin mimetics, with one qualification that I think matters. The reasoning holds for the case as described. Change the starting assumption and it does not, and the starting assumption is the part nobody states.

Effects reported at the level of appetite are the most consistently described and the most mechanistically direct for the ghrelin-receptor compounds, which is not what most people are taking them for.

I had written a reply contradicting post #11 and deleted it. Here is what survived.

Acetate versus trifluoroacetate is worth asking about for short peptides specifically, because the counter-ion is a larger proportion of the total mass at low molecular weight.

Reconstitution volumes in this family are often small enough that dead volume in the syringe is a meaningful fraction of the dose. A fixed-needle insulin syringe is worth the trouble here specifically.

Somebody will have a better source than mine, and I hope they post it.

Post #15 is right about the mechanism and I think understates the practical bit.

Sermorelin and the growth hormone releasing hormone analogues depend on a functioning pituitary response, which is the mechanistic reason they behave differently in different people rather than a dosing problem.

Ghrelin mimetics (like GHRPs) act on the ghrelin receptor directly. The effect on growth hormone secretion is similar but the mechanisms are distinct.

Coming back to post #22, because the follow-up matters more than the original answer.

Storage: these are lyophilised short peptides and are generally reasonably stable dry and considerably less so in solution. Reconstituting only what will be used within the period the supplier's own data covers is the conservative approach.

Confirming post #28 from a second method, which matters more than confirming it from a second person.
